Best British Single for One Thing and Best British Album for their debut, Up All Night were among their trophy haul.
They later thrilled fans by performing on a stage suspended over London's packed Wembley Arena.
A delighted Harry Styles expressed his joy on Twitter after his band's amazing night.
"Thank you again to BBC R1 for having us... Thank you everyone who voted! So happy to win!," he wrote.
Other artists lighting up the stage included Taylor Swift, Little Mix and Conor Maynard.
Blonde country crooner Taylor once again showcased her vampy new look, with hot pants and a slick of killer red lipstick.
Radio 1's new breakfast show host Nick Grimshaw was the man on hosting duty, holding together the star-studded evening with the help of his colleague Gemma Cairney.
It's a little princess!
